[Objective] To develop a noninvasive, rapid, reliable and cost-saving method for the identification of Pseudomonas aeruginosa. [Methods] Photoacoustic spectroscopy trace analyzer (PASTA) was adopted to analyze bacterial volatile compounds (VCs), the volatile metabolites of Pseudomonas aeruginosa and Escherichia coli. Both bacteria were cultured at 35 °C and measurements of VCs were performed at 24 h. After that the results were analyzed using the reverse thinking strategy. [Results] Volatile metabolites of Pseudomonas aeruginosa and Escherichia coli were analyzed by PASTA. A large amount of HCN was found in the VCs of Pseudomonas aeruginosa whereas none was detected in that of Escherichia coli. PASTA could be used for the identification of Pseudomonas aeruginosa. [Conclusion] Photoacoustic spectroscopy provided simple, rapid and cost-saving detection and identification of Pseudomonas aeruginosa.
